Purity and potency are related but distinct concepts in peptide testing. Purity describes the proportion of the main peptide relative to other detected substances, while potency refers to the biological or functional activity of a defined amount. A highly pure peptide can still have low potency if it is misfolded, aggregated, or chemically modified at a critical residue. Conversely, a less pure preparation may retain high activity if the impurities are inactive. Clear reporting separates these attributes and states the assay used for each.
Peptide purity specifications describe which tests define an acceptable lot and how results are reported. A certificate of analysis commonly lists a reverse-phase HPLC purity value, a mass spectrometry identity result, water content, counterion content, and residual solvent data. The specification may set a minimum area percent, such as 95% or 98%, depending on the intended use and grade. No universal threshold applies to all peptides, because sequence length, hydrophobicity, and manufacturing route influence achievable purity.
Reported purity values can differ between laboratories even for the same sample. Variations arise from column chemistry, mobile-phase composition, gradient slope, detection wavelength, injection load, and integration rules. Area percent also assumes that all species have similar response factors, which is not always true. Method validation examines specificity, linearity, accuracy, precision, limit of detection, and limit of quantitation. When comparing certificates, the method description and representative chromatogram are as important as the headline percentage.
Other methods address specific purity concerns. Amino acid analysis gives compositional data after hydrolysis, while capillary electrophoresis separates by charge-to-mass ratio. Karl Fischer titration measures residual water, and gas chromatography can detect residual solvents. Nuclear magnetic resonance can identify organic impurities but is less sensitive for trace levels. No single test covers all possible impurities, so purity testing usually combines orthogonal methods and reports the conditions used. The choice of methods is guided by the impurity classes of interest.
Reverse-phase high-performance liquid chromatography (RP-HPLC) is widely used to estimate peptide purity. It separates components by hydrophobicity on a column with a water-organic mobile phase. Ultraviolet absorbance at 214 nm or 220 nm detects peptide bonds. The main peak area as a percentage of total peak area gives a purity figure. This figure depends on column, gradient, wavelength, and how peaks are integrated, so it is method-specific rather than absolute.

== Narcoterrorism == An unwanted by-product of Bolivia's cocaine industry was the importation of Colombian-style drug violence. In the late 1980s, Colombia's Medellín Cartel reportedly wielded considerable power in Bolivia, setting prices for coca paste and cocaine and terrorizing the drug underworld with hired assassins. Furthermore, drug barons, organized into families, had established their own fiefdoms in Cochabamba, Beni, and Santa Cruz departments, using bribes and assassinations to destroy local authority. In September 1986, three members of a Bolivian scientific team were slain in the Huanchaca National Park in Santa Cruz Department shortly after their aircraft landed beside a clandestine coca-paste factory. The murders led to the discovery of the country's largest cocaine-processing installation, as well as evidence of an extensive international drug-trafficking organization consisting mostly of Colombians and Brazilians. President Paz Estenssoro fired the Bolivian police commander and deputy commander as a result of their alleged involvement. In a related action, suspected traffickers in Santa Cruz murdered an opposition deputy who was a member of the congressional commission that investigated the Huanchaca case. In the late 1980s, there were several incidents of narcoterrorism against the United States presence, the judiciary, and antidrug agents. === Vascular smooth muscle cells === Vascular smooth muscle cells play a crucial role in atherogenesis and were historically considered to be beneficial for plaque stability by forming a protective fibrous cap and synthesizing strength-giving extracellular matrix components. However, in addition to the fibrous cap, vascular smooth muscle cells also give rise to many of the cell types found within the plaque core and can modulate their phenotype to both promote and reduce plaque stability. Vascular smooth muscle cells exhibit pronounced plasticity within atherosclerotic plaque and can modify their gene expression profile to resemble various other cell types, including macrophages, myofibroblasts, mesenchymal stem cells and osteochondrocytes. Importantly, genetic lineage-tracing experiments have unequivocally shown that 40-90% of plaque-resident cells are vascular smooth muscle cell-derived, therefore, it is important to research the role of vascular smooth muscle cells in atherosclerosis to identify new therapeutic targets.

Gam-COVID-Vac is a viral vector vaccine based on two recombinant replication-defective human adenoviruses: Ad26 (serotype 26) and Ad5 (serotype 5) replicated in HEK 293 cells. The viruses contain the gene that encodes the full-length spike protein (S) of SARS-CoV-2 to stimulate an immune response. Adenoviral vectors for expression of the SARS-CoV-2 spike protein have also been used in two other COVID-19 vaccines. One is the Janssen COVID-19 vaccine, which uses the Ad26COV2 viral vector based on the human virus Ad26. For this vaccine, the cell line PER.C6 is used to replicate the vector. Another one, the Oxford–AstraZeneca COVID‑19 vaccine, uses chimpanzee adenovirus (ChAdOx1) as the vector. For both the Oxford-AstraZeneca COVID-19 and Gam-COVID-Vac vaccines the producer cells for the production of non-replicating adenoviral vectors were obtained from the HEK 293 cell line. Each dose of Gam-COVID-Vac contains (1.0 ± 0.5) × 1011 virus particles. Both Ad26 and Ad5 were modified to remove the E1 gene to prevent replication outside the HEK 293 cells. For the production of the vaccine, to propagate adenoviral vectors in which the E1 gene was deleted, HEK 293 cells are used, which express several adenoviral genes, including E1. However, although rare, homologous recombination between the inserted cellular sequence and the vector sequence can restore the replication capacity to the vector, with less than 100 replicating adenovirus particles per dose of the vaccine.
Most of the supporters also stated that the death sentence should be deployed for drug crimes due to its strong deterrent effect. Shen's death sentence was subsequently upheld after he lost his appeal on 1 March 2024. When CNN covered the topic of Singapore's death penalty and Changi Prison in October 2024, it was noted that the death penalty continued to thrive with huge public support, and there were about 40 inmates on death row as of the time when CNN covered the issue exclusively. An inmate, a former schoolteacher who introduced himself as Matthew, stated he believed in the deterrent effect of the death penalty for drug trafficking; Matthew was then serving a seven-year jail term for non-capital drug charges. However, his sentiments were not shared by the family members of death row inmates who spoke to CNN. Haslinda Ismail, whose son Muhammed Izwan bin Borhan was sentenced to death for diamorphine trafficking, stated that she was devastated over the outcome and felt resentful towards the government for not giving her son a second chance; Izwan remained on death row awaiting to be hanged as of the time of writing. Similarly, Nazeri Lajim's sister, Nazira Lajim, who was an anti-death penalty activist, stated that she was sad and angry that her brother was executed despite her push for a life sentence, and likely due to her brother's hanging in 2022, Nazira encouraged her adult children to permanently leave Singapore and emigrate to Australia.

== Medical Use == The hydrochloride salt is available as ampoules of 10 mg/ml solution for injection, 5 mg tablets, and 10 mg suppositories. It is possible that other manufacturers distribute 10 mg tablets and other concentrations of injectable nicomorphine in ampoules and multidose vials. It is used, particularly in the German-speaking countries and elsewhere in Central Europe and some other countries in Europe and the former USSR in particular, for post-operative, cancer, chronic non-malignant and other neuropathic pain. It is commonly used in patient-controlled analgesia (PCA) units. The usual starting dose is 5–10 mg given every 3–5 hours.
Common gastrointestinal side effects such as nausea, diarrhea, and vomiting are also associated with donepezil and other acetylcholinesterase inhibitors. Studies have also shown an association of donepezil use and nightmares due to increased activation of the visual cortex during sleep. Patients can be advised to complete their drug regimen in the morning to prevent occurrences of nightmares.

=== Hydrophilic interaction chromatography === Hydrophilic interaction chromatography (HILIC) is a type of liquid chromatography that uses a hydrophilic stationary phase and a high-organic mobile phase for the separation of analytes by polarity. HILIC is similar to reverse phase chromatography in its mobile phase composition, and also to normal phase chromatography, with its polar stationary phase but has distinct mechanisms. More hydrophobic analytes elute first, in an order that is generally opposite of reverse phase chromatography. In HILIC, analytes partition between the organic-rich mobile phase and a water enriched layer that forms on the surface of the polar stationary phase, with more polar analytes having stronger interactions and being retained on the column for longer. Hydrogen bonding and electrostatic interactions with the stationary phase also contribute to the separation mechanism. HILIC can be used with an isocractic buffer system, but is more commonly used with a gradient, increasing the hydrophilicity of the buffer by increasing its aqueous content so more and more hydrophilic analytes are eluted. HILIC can be used for separation of many types of analytes, but is commonly used for metabolomics studies, where it is well suited to separate polar metabolites. HILIC is orthogonal to reverse-phase chromatography, and can be combined as an orthogonal approach for more comprehensive coverage.
