Solid-phase peptide synthesis can produce truncated sequences when coupling reactions fail. Deletion peptides lack one or more internal residues, while truncation peptides end prematurely. Side reactions include aspartimide formation, oxidation of methionine, and aggregation during chain assembly. Crude synthetic peptides therefore contain target peptide plus related impurities, counterions, residual solvents, and water. Purification by preparative chromatography reduces these impurities but does not remove every closely related species, including some that differ by a single amino acid.
Quality control specifications for peptides typically include appearance, identity, purity by RP-HPLC, water content, counterion content, and residual trifluoroacetic acid. Karl Fischer titration measures water, while ion chromatography or elemental analysis can quantify counterions. Purity specifications may be set at 95% or 98% area percent, but the appropriate threshold depends on the application. For research reagents, a lower purity may be acceptable if identity is confirmed. For assays sensitive to impurities, higher purity and orthogonal testing are often required.
Purity results are only meaningful when linked to a defined sample and method. A certificate of analysis typically lists the analytical technique, column type, gradient, detection wavelength, and integration parameters. It may also report mass confirmation, water content, and counterion composition. For research peptides, laboratories often request the raw chromatogram rather than only a summary percentage. This allows independent review of baseline, peak shape, and any unresolved shoulders that might be missed by a single number.
Stability testing examines how purity changes under controlled conditions. Samples are stored at defined temperatures, such as -20 °C or -80 °C, and analyzed at intervals. Lyophilized powders are generally more stable than solutions because water promotes hydrolysis and aggregation. Repeated freeze-thaw cycles can also degrade peptides, especially those with oxidation-prone residues. Accelerated studies at elevated temperature provide useful comparisons, but they do not always predict long-term behavior at lower temperatures.
Handling practices influence measured purity. Peptides may adsorb to plastic or glass surfaces, particularly when hydrophobic or positively charged. Weighing hygroscopic powders can introduce water and alter concentration. Dissolving in appropriate solvents and using low-binding tubes can reduce losses. Each laboratory should validate its own procedures because recovery and stability vary with peptide sequence, formulation, and container material. Open questions remain about how best to standardize stability reporting across different peptide classes.
| Property | Value | Notes |
|---|---|---|
| Typical purity specification | ≥95% by RP-HPLC | Common for research-grade material; some assays require 98% or higher. |
| Water content | 5–10% w/w | Lyophilized peptides retain moisture; Karl Fischer titration measures it. |
| Counterion | Trifluoroacetate or acetate | Counterion identity affects mass balance and assay compatibility. |
| Storage temperature | -20 °C or lower | Store desiccated and protected from light; avoid repeated freeze-thaw. |
| Common impurity | Deletion or truncation peptide | Similar sequence complicates chromatographic separation. |

Gordon Flint (William Baldwin) was Adam's boss until he sold the company. When Sarah became an intern at Adam's company, she and Gordon became romantically involved, which caused friction between Adam and Sarah. Gordon is invited by Sarah to the Braverman Thanksgiving dinner, but that morning, when telling Adam he sold the company, Adam asks him not to come to the dinner. Gordon comes anyway, and at the football game that afternoon, tensions between Adam and Gordon come to a head. Mike (Kevin Alejandro), aka "Forklift Mike", drives a forklift at T&S Footwear. Mike recommends a band for Sarah to take Amber and Kelsey to see, at a club where Mike knows the man at the door. Alex (Michael B. Jordan) is Haddie's on again, off again boyfriend, whom she met while she volunteering at the food bank he currently manages. He is a recovering alcoholic and three years older than Haddie, so Haddie's parents initially disapprove of the relationship. Cory Smith (Anthony Carrigan) is Adam's new boss when Gordon sells the company. His youth and radical approach to business are disconcerting to Adam.
== History == Lipedema was first identified in the United States, at the Mayo Clinic, in 1940. Most attribute the original identification of lipedema to E. A. Hines and L. E. Wold (1951). Despite that, lipedema is barely known in the United States to physicians or to the patients who have the disease. Lipedema often is confused with obesity or lymphedema, and a significant number of patients currently diagnosed as obese are believed to have lipedema, either instead of or in addition to obesity.

Positive bottles with negative Gram stains are subcultured before being returned to the incubator, often using special culture media that promotes the growth of slow-growing organisms. It typically takes 24 to 48 hours for sufficient growth to occur on the subculture plates for definitive identification to be possible. At this point, the microbiologist will assess the appearance of the bacterial or fungal colonies and carry out tests that provide information about the metabolic and biochemical features of the organism, which permit identification to the genus or species level. For example, the catalase test can distinguish streptococci and staphylococci (two genera of Gram-positive cocci) from each other, and the coagulase test can differentiate Staphylococcus aureus, a common culprit of bloodstream infections, from the less pathogenic coagulase-negative staphylococci.
Radical transfer with a Cobalamin B12 cofactor and SAM has been shown with the few characterized radical SAM methyltransferases. Although the evidence points to radical β-methylation during bottromycin biosynthesis, it remains to be seen whether bioinformatic hypothesis and feeding studies will be supported by in vitro activity assays. The Val3Ala substitution in bottromycin D does not change the β-methylation pattern between bottromycin A2 and D because Val3 is the only valine not methylated in bottromycin A2. As such, there are still three predicted radical SAM dependent enzymes in the bottromycin D biosynthetic cluster: bstC, bstF, and bstJ. As of 2013, all published biosynthetic studies have been bioinformatic or cell-based. No biochemical assays directly demonstrating protein function have yet been published. It is likely that in vitro mechanistic studies to better elucidate the biosynthetic pathway will be forthcoming.
Collagenase 3 is an enzyme that in humans is encoded by the MMP13 gene. It is a member of the matrix metalloproteinase (MMP) family. Like most MMPs, it is secreted as an inactive pro-form. MMP-13 has a predicted molecular weight around 54 kDa. It is activated once the pro-domain is cleaved, leaving an active enzyme composed of the catalytic domain and the hemopexin-like domain PDB: 1PEX. Although the actual mechanism has not been described, the hemopexin domain participates in collagen degradation, the catalytic domain alone being particularly inefficient in collagen degradation. During embryonic development, MMP-13 is expressed in the skeleton as required for restructuring the collagen matrix for bone mineralization. In pathological situations it is highly overexpressed; this occurs in human carcinomas, rheumatoid arthritis and osteoarthritis. Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMPs are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. The protein encoded by this gene cleaves type II collagen more efficiently than types I and III. It may be involved in articular cartilage turnover and cartilage pathophysiology associated with osteoarthritis. The gene is part of a cluster of MMP genes which localize to chromosome 11q22.3.

In 2012, the American Academy of Dermatology published appropriate use criteria (AUC) on Mohs micrographic surgery in collaboration with the following organizations: American College of Mohs Surgery; American Society for Mohs Surgery; and the American Society for Dermatologic Surgery Association. More than 75 physicians contributed to the development of the Mohs surgery AUC, which were published in the Journal of the American Academy of Dermatology and Dermatologic Surgery. The Australasian College of Dermatologists, in concert with the Australian Mohs Surgery Committee, has also developed evidence based guidelines for Mohs Surgery. The Mohs procedure is a pathology sectioning method that allows for the complete examination of the surgical margin. It is different from the standard bread loafing technique of sectioning, where random samples of the surgical margin are examined. Mohs surgery is performed in four steps:

No. Purity testing measures chemical composition and does not assess biological activity, sterility, or endotoxin levels. Functional performance must be tested in the intended assay.
Water adds mass and can affect concentration calculations. A peptide labeled 95% pure may contain water and counterions that reduce the actual peptide content.
Identity can be checked by mass spectrometry, and purity by RP-HPLC. Store according to supplier instructions and retest if experimental performance changes.
It typically includes the peptide sequence, molecular mass, purity method and result, storage recommendations, and date of analysis. Raw chromatograms and mass spectra may be provided on request. The absence of method details makes a purity value difficult to interpret.
